Abstract

Generation and characterisation of monoclonal antibodies against inerolysin Lactobacillus iners is present in women vaginal microbiota. This bacterium was shown to be a key factor in the development of bacterial vaginosis and it might play a role in premature birth. L. iners is the only known Lactobacillus bacterium, which releases cholesterol-dependent cytolysin inerolysin (INY), a pore-forming toxin. This study was aimed at development of monoclonal antibodies (MAbs) against INY and MAb-based INY quantitation system. Recombinant INY was expressed in E. coli pET system. It showed an increased haemolytic activity in acidic (pH 5.4) compared to neutral (pH 6.8) solution. Following immunisation of BALB/c mice, recombinant INY was shown to induce a strong humoral immune response. After immunisation, development of IgG antibodies was observed suggesting that recombinant INY activated antigen presenting cells and T cells. Using hybridoma technology, nine MAb-producing hybridoma cell lines were generated. MAb specificity was investigated using Western Blot and ELISA methods. One MAb was cross-reactive against intermedilysin and vaginolysin. Eight out of 9 MAbs were reactive exclusively with INY. The results of a competitive ELISA allowed to classify the generated MAbs into 7 groups, directed to different immunogenic regions of INY. MAbs selected from different groups were used to develop sandwich ELISA for INY quantification. The MAb-based immunoassay was suitable for detection of recombinant INY. Generated MAbs were investigated for their ability to inhibit haemolytic activity of INY in vitro. Two out of 9 MAbs were able to inhibit INY haemolytic activity. In conclusion, the MAbs generated and described in the current study may be useful for structural and functional studies of INY as well as detection of INY in biological specimens.

Monokloninių antikūnų prieš ineroliziną kūrimas ir apibūdinimas Lactobacillus iners bakterijos yra randamos moterų lyties takų mikrobiotoje. Šios bakterijos galimai dalyvauja vystantis bakterinei vaginozei ir gali būti susijusios su priešlaikiniu gimdymu. L. iners yra vienintelės žinomos Lactobacillus šeimos bakterijos, kurios sintetina nuo cholesterolio priklausomą citoliziną ineroliziną (INY). Šio darbo metu buvo kuriami monokloniniai antikūnai (MAk) prieš INY, kurie buvo panaudoti INY kiekybinio nustatymo sistemos kūrimui. Rekombinantinis INY buvo susintetintas E. coli pET raiškos sistemoje. INY hemolizinis aktyvumas buvo didesnis rūgščioje terpėje (pH 4,5), lyginant su neutralia (pH 6,8) terpe. Imunizuojant BALB/c linijos peles rekombinantiniu INY buvo stebimas stipraus humoralinio imuninio atsako susiformavimas. Hibridomų technologijos būdu sukurtos devynios hibridomų linijos, gaminančios MAk prieš INY. Sukurti MAk buvo IgG klasės, todėl INY aktyvavo nuo T ląstęlių priklausomą imuninį atsaką. Sukurtos MAk kolekcijos specifiškumas buvo tirtas imunoblotingo ir imunofermentinės analizės (IFA) metodais. Vienas MAk rodė kryžminę reakciją prieš intermediliziną ir vaginoliziną. Aštuoni iš 9 MAk reagavo tik su INY. Atlikus konkurencinę IFA sukurti MAk buvo sugrupuoti į 7 grupes, pagal atpažįstamas skirtingas imunogeniškas sritis INY. Pasirinkus nekonkuruojančius MAk buvo sukurta dviepitopė IFA sistema kiekybiniam INY nustatymui. Sukurta sistema buvo tinkama rekombinantinio INY aptikimui mėginiuose. Buvo tiriamas MAk gebėjimas slopinti INY hemolizinį aktyvumą in vitro. Du iš 9 MAk pasižymėjo INY hemolizinio aktyvumo slopinimu. Sukurta MAk kolekcija buvo išsamiai apibūdinta įvairiais metodais ir gali būti panaudota INY struktūriniuose, funkciniuose tyrimuose ir INY aptikimui biologiniuose mėginiuose.
