Abstract

Poli-N-lactosamina (poliLacNAc) é uma estrutura fundamental nos glicoconjugados expressa especificamente em células/tecidos de forma associada ao desenvolvimento e carcinogênese. Beta-1,3-N-acetilglucosaminil transferases (β3GnTs) possuem um importante papel na síntese de poliLacNAc, entretanto a ação das β3GnTs na síntese in vivo continuam desconhecidos. A expressão de duas glicosiltransferases, β3GnT2 e β3GnT3, foram avaliadas em amostras cervicais uterinas normais e com lesões malignas (carcinoma escamoso) e prémalignas (NIC 1, 2 e 3) usando imunohistoquímica. Histoquímica com lectinas, com Phytolacca americana lectin (PWM) e Wheat germ agglutinin (WGA), também foi realizada para a análise da presença de cadeias de poliLacNAc e de N-acetilglucosamina (GlcNAc), respectivamente. β3GnT3 foi expressa em quase todas as amostras (99%) e β3GnT2 foi mais expressa nas amostras com lesões (p = 0,0093). A expressão de poliLacNAc (p = 0,0002) e GlcNAc (p = 0,0001) também se mostrou mais frequente nas amostras com lesões. Nossos dados sugerem que a β3GnT2 seja a principal enzima responsável pela síntese de cadeias de poliLacNAc na cérvice uterina e que ela possa ser utilizada como um potencial biomarcador para a presença de lesões.
