Abstract

Objectif L’hypertension systolique isolee et l’augmentation de la pression pulsee sont deux consequences de l’augmentation de la rigidite vasculaire observee chez pres de la moitie des personnes âgees de plus de 60 ans. La rigidite arterielle, causee en partie par la calcification mediale des arteres de conductance, est acceleree de 15 ans chez les diabetiques. La γ-carboxylase est une enzyme qui assure l’activation de la matrix Gla protein (MGP), une proteine anti-calcifiante, par une modification post-translationnelle. L’expression aortique de la γ-carboxylase est diminuee de facon marquee chez les rats diabetiques de type 1. L’objectif de ce projet de recherche etait d’etudier les voies de signalisation impliquees dans la modulation de l’expression de la γ-carboxylase en condition d’hyperglycemie. Materiels et methodes Des segments d’aortes de rats Wistar sains âges de 7 semaines ont ete incubes durant 24 heures avec une concentration normale (5 mmol/L) ou elevee (25 mmol/L) de glucose. Des inhibiteurs de voies de signalisation ont ete ajoutes au milieu de culture. L’expression proteique de la γ-carboxylase a ete mesuree par Western blot. Resultats Dans le modele de culture ex vivo, l’expression de la γ-carboxylase est diminuee significativement par rapport a son expression en condition normoglycemique (− 36%), tel qu’observe in vivo. L’ajout de LY333531, un inhibiteur de la voie PKCβ, a la condition hyperglycemique previent la diminution de l’expression de la γ-carboxylase. Il en est de meme lors de l’ajout de PD98059, un inhibiteur de la voie MAP kinase ERK 1/2. Conclusion Nos resultats suggerent que les voies de signalisation PKCβ et ERK 1/2 pourraient etre impliquees dans la diminution de l’expression de la γ-carboxylase en condition hyperglycemique. Cette activation des voies PKCβ et ERK 1/2 pourrait passer par la stimulation de la synthese de novo de diacylglycerol en presence d’hyperglycemie.
