Movement of RNA silencing between plant cells: is the question now behind us?
In RNA silencing, small-interfering (si)RNAs and micro (mi)RNAs are processed from double-stranded (ds)RNA by the RNaseIII Dicer [1]. siRNAs derive from long, near-perfectly base-paired dsRNAs of endogenous or exogenous origin, including viruses and transgenic hairpins used in experimental RNA interference (RNAi, Figure 1a) [2]. Endogenous miRNAs originate from DNA-encoded primary transcripts with imperfect fold-back structures; cellular targets of miRNAs include transcription factors important for cell-fate determination [3].
