Abstract

Trypanosoma brucei se basa en el empalme trans líder empalmado para generar ARN mensajeros funcionales. El corte y empalme trans une el exón SL especializado del ARN SL a los pre-ARNm y está mediado por el trans-espliceosoma, que está formado por pequeñas partículas de ribonucleoproteínas nucleares y factores no nRNP. Aunque el espliceosoma trans es esencial para la expresión génica de tripanosomátidos, no se conocen todos los factores de proteínas espliceosomales y de estos, solo unos pocos están completamente caracterizados. En este estudio, hemos caracterizado el factor de empalme del tripanosoma, SF3a60, el único componente SF3a anotado actualmente. Como se esperaba, SF3a60 etiquetado con epítopo se localiza en el núcleo del tripanosoma. SF3a60 es esencial para la viabilidad celular, pero su agotamiento parece no tener ningún efecto detectable sobre el corte y empalme en trans. Trypanosoma brucei s'appuie sur l'épissage Spliced leader trans pour générer des ARN messagers fonctionnels. L'épissage trans joint l'exon SL spécialisé de l'ARN SL aux pré-ARNm et est médié par le trans-spliceosome, qui est composé de petites particules de ribonucléoprotéines nucléaires et de facteurs non nRNP. Bien que l'épisséosome trans soit essentiel pour l'expression du gène de la trypanosomatide, tous les facteurs protéiques de l'épisséosome ne sont pas connus et parmi ceux-ci, seuls quelques-uns sont complètement caractérisés. Dans cette étude, nous avons caractérisé le facteur d'épissage du trypanosome, SF3a60, le seul composant SF3a actuellement annoté. Comme prévu, le SF3a60 marqué à l'épitope se localise dans le noyau du trypanosome. Le SF3a60 est essentiel à la viabilité cellulaire mais son épuisement semble n'avoir aucun effet détectable sur le trans-épissage. De plus, nous avons utilisé SF3a60 comme appât dans un criblage du système hybride Yeast-2 et identifié ses facteurs protéiques en interaction. Les interactions avec SF3a120, SF3a66 et SAP130 ont été confirmées par purification d'affinité en tandem et spectrométrie de masse.

Trypanosoma brucei relies on Spliced leader trans splicing to generate functional messenger RNAs. Trans splicing joins the specialized SL exon from the SL RNA to pre-mRNAs and is mediated by the trans-spliceosome, which is made up of small nuclear ribonucleoprotein particles and non-snRNP factors. Although the trans spliceosome is essential for trypanosomatid gene expression, not all spliceosomal protein factors are known and of these, only a few are completely characterized. In this study, we have characterized the trypanosome Splicing Factor, SF3a60, the only currently annotated SF3a component. As expected, epitope-tagged SF3a60 localizes in the trypanosome nucleus. SF3a60 is essential for cell viability but its depletion seem to have no detectable effect on trans-splicing. In addition, we used SF3a60 as bait in a Yeast-2-hybrid system screen and identified its interacting protein factors. The interactions with SF3a120, SF3a66 and SAP130 were confirmed by tandem affinity purification and mass spectrometry.
