Die Bedeutung eines Pro115Gln Polymorphismus im Bereich des PPARgamma2-Genes für das Auftreten von krankhafter Adipositas

Abstract

Der Peroxisomen-aktivierte-Rezeptor (PPAR) gamma2 ist ein transkriptionaler Schlüsselregulator für die Differenzierung von Adipozyten. Er kann durch eine Phosphorylierung eines Serinrestes an Position 114 inaktiviert werden. Es wurde gezeigt, daß eine Punktmutation zu einem Aminosäureaustausch an Stelle 115 (Pro115Gln) führt, der diese Phosphorylierung verhindert und sodurch die Adipozytendifferenzierung beschleunigt. Bis zu diesem Zeitpunkt konnten erst vier heterozygote Mutationsträger in einer sehr umschriebenen Studienpopulation gefunden werden, welche sich durch starke Adipositas auszeichneten (BMI 37,9 - 47,3 kg x m^2). Um die epidemiologische Relevanz der Pro115Gln Mutation bei krankhaft adipösen Personen zu evaluieren, untersuchten wir die DNA von allen Personen mit einem BMI>35 kg x m^2, welche in einer nationalen deutschen Feldstudie teilgenommen hatten. Dort fanden sich weder heterozygote noch homozygote Träger der Mutation. Wir schließen daraus, dass die Pro115Gln-Mutation des PPAR gamma2 Genes keine epidemiologische Relevanz für krankhafte Adipositas in Deutschland hat.

The peroxisome-proliferator-activated-receptor gamma2 (PPAR gamma2) is a transcriptional key regulator of adipocyte differentiation. PPAR gamma2 can be inactivated by phosphorylation of a serine residue at position 114. A point mutation leading to a amino acid exchange at position 115 (Pro115Gln) was shown to preclude serine phosphorylation and to consecutively accelerate adipocyte differentiation emphasizing the pathophysiological relevance of this mutation. So far, four markedly obese heterozygote carriers of the mutation (BMI 37.9 - 47.3 kg x m^2) have been identified in a circumscribed study population. In order to evaluate the epidemiological relevance of the Pro115Gln mutation in morbid obesity we screened the DNA of all subjects with a BMI greater than 35 kg x m^2 who hat participated in a nationwide German epidemiological field survey. There was no hómozygote or heterozygote carrier of the Pro115Gln polymorphism among them. We conclude that the mutation within the PPAR gamma2 gen has no relvant epidemiological impact on morbid obesity in Germany.
