Epigenetic DNA diagnostics of breast cancer: GLAD-PCR analysis of the DNA samples from a light fraction of the blood cells

ЭПИГЕНЕТИЧЕСКАЯ ДНК-ДИАГНОСТИКА РАКА МОЛОЧНОЙ ЖЕЛЕЗЫ: GLAD-ПЦР АНАЛИЗ ДНК ЛЕГКОЙ ФРАКЦИИ КЛЕТОК КРОВИ

Abstract

Эпигенетическая ПЦР тест-система «EpiGene BC» позволяет определять РМЖ путем GLAD-ПЦР-анализа ряда сайтов RCGY в препаратах ДНК, связанной с клетками крови. С использованием данной тест-системы определяли уровень метилирования двух сайтов RCGY в препаратах ДНК, выделенной из легкой фракции клеток крови, полученной от доноров и больных РМЖ на разных стадиях заболевания. Исследование включало а) получение легкой фракции клеток крови, б) выделение ДНК из легкой фракции клеток крови и определение ее концентрации методом ПЦР в реальном времени в) установление концентрации сайтов R(5mC)GY в генах 1 и 2 в полученных препаратах ДНК методом GLAD-ПЦР-анализа и определение доли метилированных сайтов от общей концентрации ДНК. Показано, что диагноз РМЖ может быть исключен, если в препарате ДНК легкой фракции клеток крови уровень метилирования сайта RCGY в гене 1 по данным GLAD-ПЦР-анализа составляет не менее 80%. При низком уровне метилирования сайта RCGY в гене 1 (менее 80%) диагноз РМЖ также может быть исключен, если уровень метилирования сайта RCGY в гене 2 достоверно ниже 3,7%. Метод GLAD-ПЦР-анализа не требует сложного и дорогостоящего оборудования и реактивов, а само исследование может проводиться в стандартной ПЦР-лаборатории в ходе ежегодных профилактических обследований и анализа крови.

Epigenetic PCR test-system “EpiGen BC” for diagnostics of breast cancer is based on determination of methylation of several sites RCGY in samples of DNA bound to the blood cells. We have used this test-system to determine methylation of two RCGY sites in DNA preparations isolated from the light fraction of the blood cells of donors and BC patients at different stages of disease. The study includes: a) preparation of the light fraction of the blood cells, b) isolation of DNA from the light fraction of the blood cells and determination of DNA concentration with real-time PCR and c) determination of concentration of RCGY site in genes 1 and 2 followed by calculation of their percentage in a total DNA. We have shown that the BC diagnosis may be denied if GLAD PCR assay of RCGY site in gene 1 has shown a percentage of its methylation 80% and more. In case of low methylation of RCGY site in gene 1 (less than 80%) the BC diagnosis may be denied if methylation of RCGY site in gene 2 is definitely less than 3,7%. GLAD PCR assay doesn’t require special and expensive equipment. The study can be carried out in a standard PCR laboratory when a blood analysis is taken place.
