Interest of rabbit induced pluripotent stem cells for species preservation –

Abstract

Pour maintenir la biodiversité d’une espèce donnée, il est essentiel de développer les techniques de cryoconservation permettant de conserver à long terme les ressources génétiques de cette espèce. Pour cela, on peut conserver des embryons, des gamètes mâles ou femelles, ou encore des tissus somatiques. Les techniques de prélèvement et de cryoconservation des embryons et du sperme sont efficaces et couramment utilisées chez le lapin, mais elles ne sont pas applicables à tous les individus. Afin d’élargir le nombre d’échantillons conservés, il est donc souhaitable de pouvoir congeler des tissus somatiques qui peuvent être facilement prélevés sur le terrain sans préparation préalable de l’animal. Les cellules somatiques ainsi conservées pourront ensuite être reprogrammées en cellules souches pluripotentes induites (ou iPSC), afin de produire des animaux chimères. Nous ferons le point sur l'état des recherches actuelles sur les iPSC de lapin et leur intérêt pour la conservation de cette espèce

Developing cryobanking techniques is essential to maintain the biodiversity of a given species and the long-term preservation of its genetic resources. For that purpose, we can preserve embryos, male or female gametes, or somatic tissues. Sampling and cryogenic preservation of embryos and sperm are effective and commonly used in rabbits, but they are not applicable to all animals. Consequently, to widen the number of preserved samples, it is necessary to freeze somatic tissues such as fragments of skin or blood cells. These somatic cells could afterward be reprogrammed in pluripotent stem cells (or iPSCs) in order to produce animal chimeara. This paper will review the current state of rabbit iPSC researches and their interest for species preservation.
